The present invention provides a genetic tool for producing a recombinant microorganism by inserting, substituting, deletion or otherwise manipulating a nucleic acid sequence in a microorganism. Significantly, the invention utilizes homologous recombination, a genetic recombination type, in which the nucleotide sequence is exchanged between two similar or identical DNA molecules. Three homologous recombination events are involved in the present invention, which is referred to as the "triple crossing" method. |
