abstract |
The present invention relates to a cross-neutralizing antibody comprising at least one multispecific binding site that binds to the alpha-toxin (Hla) of Staphylococcus aureus and to at least one two-component toxin, Wherein the antibody comprises at least three complementarity determining regions (CDR1 to CDR3) of the antibody heavy chain variable region (VH), wherein A) said antibody comprises: a) a CDR1 comprising or consisting of the amino acid sequence YSISSGMGWG (SEQ ID NO: 1); And b) a CDR2 comprising or consisting of the amino acid sequence SIDQRGSTYYNPSLKS (SEQ ID NO: 2); And c) a CDR3 comprising or consisting of the amino acid sequence ARDAGHGVDMDV (SEQ ID NO: 3); Or B) said antibody comprises: a) a parent CDR1 comprising the amino acid sequence of SEQ ID NO: 1; And b) a parent CDR2 consisting of the amino acid sequence of SEQ ID NO: 2; And c) a parent CDR3 consisting of the amino acid sequence of SEQ ID NO: 3; Wherein said functionally active CDR variant comprises or consists of an amino acid sequence comprising at least one point mutation in said parent CDR sequence and having at least 60% sequence identity to said parent CDR sequence. The present invention is also directed to such a cross-neutralizing antibody as a functionally active variant antibody of a parent antibody comprising a VH amino acid sequence of SEQ ID NO: 20 and a multispecific binding site of a VL amino acid sequence of SEQ ID NO: 39, antibody comprises at least one point mutations in any region of the framework regions (FR) or a constant domain, or SEQ ID NO: 20 and SEQ ID NO: complementarity determining regions (CDR1 to CDR6) of any of the sequences of the 39 and 10 -8 K, preferably less than 10 < -9 > M. |
