abstract |
The present invention relates to a method for adding sialic acid to an oligosaccharide of a glycoprotein using a sialic acid transferase having improved efficiency. More specifically, the present invention relates to a method of adding sialic acid to a glycoprotein, Wherein the reaction is carried out while removing CMP derived from CMP-Neu5Ac in the step of adding sialic acid to the sugar chain of CMP-Neu5Ac. The CMP removal in the present invention can be accomplished by converting CMP to cytidine through the dephosphorylation of CMP or by converting it to CDP, CTP or CMP-Neu5Ac by adding phosphoric acid to CMP, The efficiency of the addition reaction can be increased. In addition, since the sialic acid transferase is derived from bacteria, the sialic acid transferase is more economical and can be mass-produced than conventional sialic acid transferase, so it can be easily used. |
