abstract Provided in the present invention is a method to detect a rheumatism marker through a surface enhanced Raman scattering (SERS) based immunoassay comprising: a step of preparing a test sample solution expected to contain an autoantibody, an anti-cyclic citrullinated peptide (CCP) antibody; a step of preparing magnetic particles fixated with a cyclic citrullinated peptide (CCP) specifically combined with the anti-CCP antibody; a step of preparing a nanoprobe combined with a secondary antibody which is specifically combined with the anti-CCP antibody and a Raman reporter molecule; a step of forming a first immune complex through an immune reaction by CCP of the magnetic particles and the anti-CCP antibody by adding the magnetic particles in the test sample solution; a step of forming a sandwich immune complex through a combination of the secondary antibody of the nanoprobe and the anti-CCP antibody of the first immune complex; a step of separating the sandwich complex using magnetism; a step of radiating a laser beam to the separated sandwich immune complex; and a step of confirming an existence of the autoantibody, the anti-CCP antibody by measuring surface-enhanced Raman scattering (SERS) signals after radiating with the laser beam.
