abstract |
The methods and compositions specifically process samples for sequencing applications. The bead composition for various libraries of beads attached to a plurality of oligonucleotides contains a barcode. The method includes a method of producing a library of barcoded beads, for example, a method of combining a bead with a sample using a microfluidic device. The first compartment includes a plurality of nucleic acid barcode molecules associated therewith, and the nucleic acid barcode molecule may comprise the same nucleic acid barcode sequence. The first compartment may be co-compartmentalized together with the components of the sample material into the second compartment, and then the bar code molecules may be liberated from the first compartment to the second compartment. The liberated barcode molecule is attached to a component of the sample material or a fragment thereof in the second compartment. The first fraction may comprise up to 10000+ barcode molecules associated therewith, having the same barcode sequence. |
