abstract The present invention relates to antisense oligonucleotide (ASO) compositions and, in particular, to compositions and methods for cytoplasmic delivery of antisense oligonucleotides (ASO). Partial single stranded and partially double stranded hybrid ASOs are provided, which hybridize to form double stranded regions that can non-covalently bind to the nucleic acid-binding protein region. In this way, an ASO :: protein complex can be produced which facilitates delivery of antisense DNA into the target cell. These complexes can be used to down-regulate gene expression in cells.
