abstract The present invention relates to hydrogel-in-oil emulsions containing cells, an in vitro compartmentalization (IVC) method, and an ultra-high throughput screening (uHTS) method for novel enzymes. The present invention can prevent the diffusion of a product generated by a reaction between cells and a substrate in hydrogel to enhance the fluorescence intensity, thereby being able to be utilized for a fluorescence-activated cell sorting (FACS) analysis. According to the present invention, the emulsions prepared by using the IVC method are stable and are able to be stored for an extended period. The emulsions can be used to culture cells and enable a single cell to proliferate to two or three cells.
