The present invention provides a method for making liposomes, lipid disks, and other lipid nanoparticles using multi-port manifolds, wherein a lipid solution stream containing an organic solvent is mixed with two or more aqueous (e.g., buffer) streams. In some aspects, at least some of the streams of lipid solution and aqueous solution are not directly opposite to each other. Thus, the process does not require dilution of the organic solvent as an additional step. In some embodiments, one of the solutions may also contain an active pharmaceutical ingredient (API). The present invention provides a preferred method of liposomes prepared with different lipid formulations and different loading amounts. The particle size, shape, and manufacturing scale can be adjusted by changing the number and port size of the manifold ports and by selecting the flow rate or flow rate of the lipid solution and aqueous solution. |
