abstract The present invention provides a bioreactor for large-mammal lung tissue. The bioreactor is capable of hydraulically driven negative pressure and positive pressure perfusion and ventilation. Perfusion and ventilation are delivered at physiological rates and are easily controllable. In one embodiment, the bioreactor includes a support scaffold for supporting the size of the large-mammal lung tissue. In another embodiment, the bioreactor includes a pleural pouch that provides a small isolated fluid chamber that surrounds the engineered lungs and minimizes the amount of culture medium required. The present invention also provides in vitro models for investigating the function of test agents and compositions and methods for alleviating lung defects in large-mammals.
