In certain embodiments, hydantoin compounds effective to inhibit BACE activity against APP are provided herein. Without being bound by a particular theory, it is believed that the activity of the hydantoins identified herein appears to be associated with binding to BACE and / or APP, particularly when the moiety forms a BACE / APP complex. Thus, it is believed that the compounds described herein represent a new class of compounds referred to herein as APP binding-BACE inhibitors (ABBI) and provide a new mechanism to control APP processing. The hydantoins described herein appear to exhibit improved brain permeability and functional BACE inhibition. |
