abstract In the peripheral blood-derived lymphocyte cells, about 50% of the inexperienced T cells are occupied before the activation culture, but the rate is decreased by the activation culture. On the other hand, the share of memory T cells, which was less than half of that before the start of culture, increased to 95% due to the activation culture, but gradually decreased when the culture was continued again. It is an object of the present invention to provide a method for massively preparing a CD45RO positive T cell group in a simple and rapid manner without using an antibody or the like, a pharmaceutical composition containing a memory T cell group prepared by the above method as a main component, and the like do. The present invention detects activated lymphocyte cells activated and cultured and shows that the percentage of the memory T cells in the lymphocyte cell group has reached a desirable value with the index of the cell density of the lymphocyte cell group in culture exceeding a predetermined threshold value, The cell group is harvested, cryopreserved, and the cryopreserved cells are thawed, cultured, and proliferated to thereby produce a large number of lymphocyte cell groups containing memory T cells as a main component.
