titleOfInvention | Novel DHFR-deficient CHO cell line transfected by a vector which over-expresses Bcl-2 and Beclin-1 protein, and a method for preparation thereof |
abstract | The present invention relates to a novel CHO host cell line transformed to simultaneously overexpress Bcl-2, an anticipated gene, and Beclin-1, a child-acting pathway gene, and lacking a dhfr (dihydrofolate reductase) gene and a method for producing the same. The transgenic CHO cell line of the present invention can inhibit cell proliferation due to overexpression of Bcl-2, which is an anti-prodrug protein, and induce cell death due to overexpression of Beclin-1, And it was confirmed that the induced pathway could inhibit cell death, and it was confirmed that cell death was more effectively inhibited than overexpression of Bcl-2, which is a conventional cell death suppression method, By confirming that the cell death induced by stress is inhibited, a therapeutic protein can be advantageously used for production. |
