titleOfInvention | IGF-I Isolation Method from Cow Colostrum |
abstract | The present invention provides a method for isolating insulin-like growth factor 1 (IGF-I) from mammalian milk comprising the steps of: (a) separating mammalian milk into whey and casein; (b) acidifying the whey of step (a) to pH 1-4 and then centrifuging to recover a fraction containing a large amount of IGF-I; (c) fractionating a fraction containing a large amount of IGF-I in the step (b) by ion exchange chromatography using an anion exchange resin to obtain a fraction containing a large amount of a substance corresponding to the molecular weight of IGF- Separating; (d) separating a fraction containing a large amount of a substance corresponding to the molecular weight of IGF-I of the step (c) by using a gel filtration chromatography; And (e) passing a fraction containing a large amount of IGF-I in step (d) through a filtration membrane of 7000-13000 Da to recover a fraction of 7000-13000 Da or less, and concentrating the filtrate to 700-1300 Da to recover IGF- step. The method of the present invention provides the advantage that the separation and purification process is significantly simpler than the previously reported IGF-I separation and purification method (Francis et al., 1986). In addition, the method of the present invention has an advantage that the content of IGF-I recovery can be increased three times or more as compared with the previously reported IGF-I separation and purification method. |
