abstract |
The present invention provides a) a polynucleotide that grows a culture of Gram negative host cells transformed with a polynucleotide encoding bacterial toxoid and plasma membrane periphery signal sequences in fermentation media, or encodes bacterial toxoid and plasma membrane periphery signal sequences. Providing a Gram negative host cell comprising bacterial toxoid transformed with and expressed in the periplasm of the plasma membrane; a (i)) inducing the expression of a bacterial toxoid; b) maturing the host cell, wherein the maturation step comprises the steps of: I) subjecting the host cell to pH shock; II) incubating host cells without feed addition; And / or III) treating the host cell at a temperature below −20 ° C .; And c) extracting the bacterial toxoid from the host cell, wherein the extraction process comprises an osmotic shock. |
