abstract Chemical treatment containing target biomolecules of interest, such as mAb, mammalian cell cultures, or bacterial cell cultures, using a primary clarification deep filtration without the use of a primary clarification centrifugation step or a primary clarification tangential flow microfiltration step. To purify the feed, including the flocculated flocculated feed. The primary clarification depth filtration device contains a porous depth filter having a graded porous layer of varying porosity. The primary clarification depth filtration apparatus provides a fluid feed of about 10 liters / m 2 , including agglomerated cell lysate having a particle size distribution of about 0.5 μm to 200 μm and a chemically treated agglomerated feed containing colloidal particulates. Filter at a flow rate from / hr to about 100 liters / m 2 / hr. Kits and methods of using and preparing the same are also provided.
