abstract The present invention relates to the detection of target nucleic acid sequences by PCE (PTO Cleavage and Extension-Dependent Cleavage) assays. The present invention is characterized in that a cleavage site that is cleaved by a nucleic acid cleavage enzyme is generated on an extended dimer formed according to the presence or absence of a target nucleic acid sequence. The present invention detects the occurrence of extended dimeric cleavage, thereby determining the presence of the target nucleic acid sequence.
