abstract | PURPOSE: Detecting method of Nucleotide variation is provided to easily realize the reaction condition by locating the region to distinguish the nucleotide variation to the 5'- distal point and accurately distinguish the difference of one nucleotide sequence as distinguishing the nucleotide variation through the localized parts. CONSTITUTION: A method of detecting nucleotide variation within the target nucleic acid sequence comprises (a) a step of mixing the target nucleic acid with upstream oligonucleotide and Probing Oligonucleotide (PO); (b) a step of cutting the PO by letting enzyme having 5' nuclease activity contact to the outcome of the step (a) under the condition for cutting the PO; and (c) a step of detecting signal. (c) The step of detecting signal of computer is included. The detection of the signal exhibits the presence of the nucleotide variation. |
