abstract | The present invention provides a composition comprising (a) a centromere sequence; (b) a sequence homologous to an upstream flanking sequence of the gene of interest (UP fragment); (c) (i) ScTDH3 ( Saccharomyces cerevisiae an expression module comprising a promoter of glyceraldehyde-3-phosphate dehydrogenase 3), (ii) a multiple cloning site (MCS), and (iii) a terminator of ScGAL7; And (d) (i) a selection marker; And (ii) a rescue module (loxP-selection marker-loxP) comprising a loxP sequence linked to both ends of the selection marker; (e) The present invention relates to a yeast expression vector for chromosome integration comprising a homologous sequence (down fragment) to a downstream flanking sequence of a gene of interest and a transformation method using the same. The transformation method using the vector of the present invention not only stably expresses a target gene in a cell even after generations, but also additionally includes a flanking sequence of the target gene fragment to further detect one or more target genes. As it is possible to induce continuous transformation, it can be very usefully applied to the industrial production of desired products (biomass). |
