Described herein are immunostimulatory compositions and methods comprising an ITIM motif-containing DC immunoreceptor (DCIR) to mediate strong cross presentation. We evaluated human CD8 + T cell responses generated by targeting antigens to dendritic cells (DCs) through various lectin receptors. Single exposure to low doses of anti-DCIR-antigen conjugates resulted in antigen-specific CD8 + T cell immunity by ex vivo DCs, skin-separated Langerhans cells and all human DC subsets including blood mDCs and pDCs.
