abstract |
Disclosed are 'hot start' RNAse H enzyme compositions that are reversibly modified for improved CATACLEAVE ™ probe detection of nucleic acid sequences in test samples. An important feature of the enzyme composition is the ability to modulate the catalytic activity of RNAse H during reverse transcription-PCR reactions. Thus, RNAse H activity can be initially inhibited to minimize degradation of RNA: DNA primer heteroduplex prior to reverse transcription. After cDNA synthesis is complete, CATACLEAVE ™ induces RNAse H activity to anneal to target DNA sequences in reverse transcriptase-PCR products Promote cleavage and fluorescence detection of the probe. The inducible RNAse H enzyme is a one-step reverse transcriptase CATACLEAVE ™ in a single reaction mixture. It is suitable for high-speed mass analysis requiring PCR. |
