The present invention relates to a method for detecting a probing and tagging oligonucleotide (PTO) hybridized with a target nucleic acid sequence and cleaving a fragment to hybridize to a capturing and template oligonucleotide (CTO) to form an extended duplex, Thereby detecting the target nucleic acid sequence. The extended dimer provides a signal (generation, increase, extinction, or reduction of the signal) from the indicia indicating the presence of the extended dimer and has an adjustable Tm value, which can be applied to the detection of the presence of the target nucleic acid sequence.
