abstract It is an object of the present invention to provide an amplification method that suppresses amplification by annealing of a wrong primer. In amplification of a target sequence comprising a target site exhibiting a polymorph, primers X1 and X2 are used. X1 has sequence A1 'and sequence E1, A1' is a sequence complementary to partial sequence A1 in the said template nucleic acid, and the 3 'area | region is based on the preparation of the said target site in 5' area | region of A1. A primer having a base x1 'complementary to monobasic x1, E1 is a non-complementary sequence to the partial sequence B1 adjacent to the 3' end of A1 in the template nucleic acid, and is bound to the 5 'end of A1'. Shall be. X2 has sequence A2 ', A2' is a sequence complementary to the partial sequence A2 in the said template nucleic acid, and the 3 'area | region is the 2nd base x2 of the said target site | part in the 5' area | region of A2. It is set as the primer which has base x2 'complementary to. X1 and X2 together have a base complementary to the target site in the 3 'region. These primers can prevent misamplification of the target sequence having the second base x2 in the case of only the template having the first base x1. Thereby, false positives can be suppressed with respect to the polymorph of 2nd base x2.
