abstract | It is an object of the present invention to provide an amplification method that suppresses amplification by annealing of a wrong primer. In amplification of a target sequence comprising a target site exhibiting a polymorph, primers X1 and X2 are used. X1 has sequence A1 'and sequence E1, A1' is a sequence complementary to partial sequence A1 in the said template nucleic acid, and the 3 'area | region is based on the preparation of the said target site in 5' area | region of A1. A primer having a base x1 'complementary to monobasic x1, E1 is a non-complementary sequence to the partial sequence B1 adjacent to the 3' end of A1 in the template nucleic acid, and is bound to the 5 'end of A1'.
