Single-molecule detection systems and methods of detecting single-molecule individuals or sequencing nucleic acids are provided. The single-molecular detection system includes a movable optical coupler 100, a waveguide 110 and a photo detector 102. Waveguide 110 includes a core layer 112 and a first cladding layer 114, wherein at least one adapter site 104 for the movable optical coupler 100 is formed in at least the first cladding layer 114. . Detection systems are used to detect single-molecule individuals or to sequence nucleic acids. |
