Cloning, Yeast Expression, Purification and Biological Activity of the Extension Region of Soybean 7G Globulin Alpha´ Subunits Associated with HepP2 Cell Cholesterol Homeostasis |
abstract |
A truncated form of the α 'chain (eα'), soybean 7S globulin, which is active in regulating cholesterol and triglyceride homeostasis in in vitro and in vivo models, was cloned and expressed in yeast Peachia pastoris. The recombinant polypeptide consists of 142 amino acid residues from the N-terminal side and comprises the N-terminal extension region of the soybean alpha 'subunit. The eα 'polypeptide was purified by conventional biochemical techniques, and the possibility of modulating the activity of LDL-receptors was evaluated by monitoring the uptake and degradation of labeled LDL in human liver cancer cell line (Hep G2). |
