The present invention relates to a method of culturing cells, preferably E1-immortalized HER cells, more preferably PER.C6 cells, in suspension in a cell culture medium in a reactor, wherein the cells comprise a biological material, preferably an antibody. One or more cell culture medium components are supplied to the cell culture, the cell culture comprising cells, biological material and cell culture medium is circulated on a separation system, and the separation system is biologically derived from a material having a lower molecular weight than the biological material. The material is separated and the biological material is retained in the reactor or fed back to the reactor. Preferably, a portion of the material whose molecular weight is lower than the biological material is continuously removed from the cell culture. |
