abstract Provides a high temperature-stable, highly purified crosslinked (optionally ≥70% β-β bound) tetrameric hemoglobin with high efficiency oxygen delivery suitable for use in mammals without causing kidney damage and vasoconstriction do. The dimeric form of hemoglobin is altered and a purification process is performed on red blood cells from whole blood. Regulated hypotonic lysis in an instant cytolysis device prevents lysis of white blood cells. Nucleic acid and phospholipid impurities from leukocytes are not detected. Blocking by the sulfhydryl reagent of the reactive sulfhydryl group is performed in an oxygenated environment. Perfusion column chromatography removes different plasma protein impurities. N-acetyl cysteine is added to the crosslinked tetrameric hemoglobin to maintain low levels of met-hemoglobin. The stabilized hemoglobin is preserved in an infusion bag with aluminum overpack to prevent the formation of inert meth-hemoglobin from oxygen invasion. The product is used for tissue oxygen supply and cancer treatment.
