titleOfInvention | Means for purifying coagulant proteins and methods of performing the same |
abstract | The present invention relates to an affinity matrix that selectively binds to a coagulant protein, wherein the nucleic acid aptamers that specifically bind to the coagulation protein comprise an immobilized solid substrate material. In some embodiments, the nucleic acid aptamer consists of deoxyribonucleic aptamers. It is also an object of the present invention a method of immobilizing a coagulant protein on a substrate, comprising contacting a sample comprising the coagulant protein with an affinity substrate. The invention also relates to a method for purifying coagulant protein, comprising the following steps: a) contacting a sample comprising a coagulant protein with an affinity matrix as defined above to form a complex of (i) a nucleic acid aptamer immobilized on an affinity matrix and (ii) a coagulant protein, and b) releasing the protein from the complex formed in step a), and c) recovering the coagulated protein in purified form. The invention also relates to a purified composition of recombinant human coagulant protein comprising at least 99.9% by weight of recombinant human coagulant protein and substantially free of non-human protein. |
