abstract The present invention relates to in vivo imaging, in particular to in vivo imaging of peripheral benzodiazepines receptor (PBR). Tetracyclic indole in vivo imaging agents are provided that bind to PBR with high affinity, have good brain uptake after administration, and preferentially bind to tissues expressing high levels of PBR. The present invention also provides precursor compounds useful in the synthesis of the in vivo imaging agents of the invention, methods of synthesizing the in vivo imaging agents comprising the use of the precursor compounds, and kits for performing the methods. Cassettes for automated synthesis of in vivo imaging agents are also provided. The present invention also provides a radiopharmaceutical composition comprising the in vivo imaging agent of the invention, and a method of using said in vivo imaging agent.
