The application provides an isolated monoclonal antibody, in particular a human monoclonal antibody, that specifically binds LAG-3 with high affinity. Preferably, the antibody binds to human LAG-3. In certain embodiments, the antibody binds to human and monkey LAG-3 but not to mouse LAG-3. The present invention provides anti-LAG-3 antibodies capable of inhibiting the binding of LAG-3 to MHC class II molecules and stimulating antigen-specific T cell responses. Also provided are nucleic acid molecules encoding expression antibodies, expression vectors, host cells, and methods of expressing antibodies of the invention. Also provided are immunoconjugates, bispecific molecules and pharmaceutical compositions comprising the antibodies of the invention. The present disclosure also provides methods of detecting LAG-3 using anti-LAG-3 antibodies of the invention, and methods of treating stimulatory immune responses. Also provided is a combination therapy in which the anti-LAG-3 antibody is co-administered with at least one additional immunostimulatory antibody. |
