abstract | The present invention relates to a method and kit for detecting a target nucleic acid sequence in a DNA or nucleic acid mixture using a 5′-cleavage and 3′-extension reaction of a target hybridization and detection primer (THD primer). The present invention is characterized by using a THD primer having a dual function of providing a signal capable of detecting a target nucleic acid sequence as well as synthesizing a base sequence complementary to the target nucleic acid sequence. When the THD primers of the present invention are used in real-time PCR, not only amplification of the target nucleic acid sequence accompanied by an extension reaction of the reverse primer as a primer, but also without additional labeled probes as in the conventional manner, The 5'-cleavage reaction of the template dependent nucleic acid polymerase with 'to 3' nuclease activity can provide a detection signal of the target nucleic acid sequence. Accordingly, the present invention can achieve the object of amplifying the target nucleic acid sequence and real-time detection of the target nucleic acid sequence even using only the THD primer and the reverse primer. |
