abstract | An object of the present invention is to provide a method for efficiently producing a copolyester comprising 3HB and LA by microbial fermentation using sugar as a raw material. It is a manufacturing method of the copolyester containing hydroxybutyric acid and lactic acid, 1) a protein catalyzing the reaction of CoA transfer to propionic acid and / or lactic acid, a protein catalyzing the reaction of acetoacetyl CoA formation from two molecules of acetyl CoA, a protein catalyzing the reduction reaction of acetoacetyl CoA, and a A method of culturing a recombinant microorganism having a protein consisting of the amino acid sequence of c) or b) and catalyzing a synthesis reaction of polyhydroxyalkanoic acid in a medium containing a carbon source: a) an amino acid sequence in which at least one of the 130, 325, 477, and 481 amino acids of the amino acid sequence represented by SEQ ID NO: 2 is substituted with another amino acid; b) in the protein as defined in a), additionally an amino acid sequence wherein at least one or more amino acids other than 130, 325, 477 and 481 are deleted or substituted, or at least one amino acid residue is inserted, and 2) A method for producing the copolyester comprising the step of recovering the copolyester from the culture of step 1). According to the production method of the present invention, a copolyester comprising 3HB and LA can be efficiently produced using an inexpensive carbon source, and the production cost of biodegradable plastics can be lowered. |
