abstract The present invention provides a variation detection method that enables variation detection with excellent sensitivity and reliability in one reaction system. Using the primers (Xmt) and (Xwt), the target nucleic acid sequence whose base for detection is a variant is amplified with an amplification efficiency higher than that of the target nucleic acid sequence whose base for detection is a normal type. (Xmt) is a primer having a base complementary to a region containing a mutant base in the template nucleic acid and a base complementary to the variant base in the 3 'region, and (Xwt) is a region containing a normal base in the template nucleic acid Primer having a base complementary to and complementary to the normal base in the 3 'region. It is preferable that the amplification efficiency by (Xmt) with respect to the variant template nucleic acid is higher than the amplification efficiency by (Xwt) with respect to the normal template nucleic acid. And the signal value which shows the melting state of the hybridization body of the obtained amplification product and a probe is measured, and the presence or absence of the variation of the target base site is determined from the signal value change accompanying a temperature change.
