titleOfInvention | Method for Enhancing Taxane Production by Cell Culture of Taxus Species |
abstract | The present invention includes all known Taxus species, for example, brevifolia, canadensis, cuspidata, baccata, globosa, florina ( methods for producing Taxol, Bacatin III and other Taxol-like compounds in very high yields from floridana, wallichiana, media and chiensis. Certain modifications of culture conditions (ie, medium composition and mode of operation) have been found to increase the yield of various taxanes from cell cultures of all Taxus species. Particularly preferred enhancers include silver ions or silver complexes, jasmonic acid (particularly methyl esters thereof), auxin-related growth regulators and inhibitors of the phenylpropanoid pathway (eg 3,4-methylenedioxy-6-nitrocinnamic acid) have. These enhancers can be used alone or in combination with each other or with different yield increasing conditions. While the yield of taxanes from plant cell cultures of Taxus chinensis is particularly increased using one or more of these conditions, the yield of taxanes for Taxus species has been found to benefit from the use of these conditions. |
