abstract | The present invention relates not only to methods for increasing the titer of a protein of interest in cells, but also to improved production and purification of optimized biomolecules whose one component is domain C H 3. In biomolecules, C-terminal amino acid (s), for example C-terminal lysine, are frequently cleaved. For example, typically incomplete treatment of heavy chains of antibodies leads to product heterogeneity. To avoid this product heterogeneity, the corresponding codons of the C-terminal lysine of the antibody heavy chain are deleted by recombinant DNA technology. These optimized antibodies induce higher product titers than in the wild type. In addition, these optimized antibodies are advantageous during the purification process by having improved elution characteristics as a result of reduced charge heterogeneity. |
