abstract |
The present invention provides a method of engineering a protein such that the performance of the protein is optimized under specific environmental conditions of interest. In some embodiments, the present invention provides a method of engineering an enzyme to optimize the catalytic activity of the enzyme under certain environmental conditions. In some preferred embodiments, the present invention provides a method of engineering an enzyme such that the catalytic activity and / or stability of the enzyme under opposing environmental conditions is optimized. In some preferred embodiments, the present invention provides a method of engineering an enzyme such that the storage stability of the enzyme is optimized under particularly opposing environmental conditions. In some preferred embodiments, the present invention provides a net surface charge and / or an enzyme (eg metalloprotease) such that an enzyme variant is obtained that exhibits improved performance and / or stability in detergent formulations as compared to the starting or parent enzyme.
