abstract | The present invention relates to RNA oligonucleotides or RNA oligonucleotide complexes (also referred to herein as RNA complexes) comprising one or more, optionally more than one, hydroxymethyl substituted nucleotide monomer (s). Nucleotide monomers comprising a hydroxymethyl group (which may be unsubstituted, eg O-substituted with a linking group, or optionally converted to an aminomethyl group which is substituted or linked) are considered hydroxymethyl substituted nucleotide monomers. The hydroxymethyl group does not participate in the formation of internucleotide bonds and is not the hydroxymethyl group (including the 5'-hydroxy group) of the native RNA monomer. RNA complexes of the invention may be useful for therapeutic, diagnostic or research use. Such complexes include short interfering RNA complexes (siRNA double strands), including antisense strands and continuous or discontinuous passenger strands ('sense strands') and capable of regulating gene expression. One or more of the hydroxymethyl substituted nucleotide monomer (s) of the invention, which may be located at 3'-terminus, 5'-terminus or internally, at least one such strand, possibly more than one such strand.
