abstract |
The present invention is followed by precipitation with ammonium sulfate, hydrophobic interaction chromatography, cation exchange chromatography, and anion chromatography followed by Clostridium histolyticum collagenase types I and types from complex mixtures. A method of purifying a II protein is provided. Conditions are provided to cause stabilized, partially purified formulations even after the precipitation step. The method of the present invention removes other proteolytic activity quickly and efficiently. The formulations according to the present invention provide pure and intact collagenase type I and type II proteins that are unexpectedly enzymatically active. The present invention also provides a blend of two isolated proteins. The present invention further provides for the use of purified collagenase proteins or blends thereof for treating tissue samples in vitro. |
