abstract | Methods and compositions are provided for the development of effective cancer therapies using mitotic inhibitors with limited general toxicity to normal non-cancer cells and tissues. The methods and compositions utilize cytotoxic compounds consisting of cell-binding agents (eg antibodies) conjugated to anti-mitotic compounds (eg maytansinoids). The invention provides that the therapeutic effect is not due to indirect cell killing by ADCC and / or CDC by substantially inducing antibody-dependent cell-mediated cytotoxicity (ADCC) and / or complement dependent cytotoxicity (CDC). Further provided are antibodies that can ensure that they are primarily mediated by the anti-mitotic component of the cytotoxic compound. Antibodies of the invention can distinguish polypeptide antigens that are more highly expressed in proliferative cancer cells as compared to proliferative non-cancer cells. |
