The present invention describes a method for identifying, isolating and producing a hyperimmune serum-reactive antigen from a particular pathogen, tumor, allergen, or tissue or host susceptible to autoimmunity, which antigen is used as a vaccine in humans or certain animals. And the method is characterized by the following steps:n n n Providing an antibody preparation from animal plasma, or human plasma or individual serum, comprising an antibody against a particular pathogen, tumor, allergen, or tissue or host that is susceptible to autoimmunity;n n n Providing at least one expression library of tissue or host that is susceptible to particular pathogens, tumors, allergens, or autoimmunity;n n n Selecting at least one expression library using antibody preparation;n n n Identifying antigens which bind to antibodies in the antibody preparation during the selection process;n n n Screening the identified antigens with individual antibody preparations derived from individual sera of an individual comprising an antibody against a particular pathogen, tumor, allergen, or tissue or host that is susceptible to autoimmunity;n n n Identifying the hyperimmune serum-reactive antigen portion of the identified antigen, wherein the hyperimmune serum-reactive antigen binds to the relevant portion of the individual antibody preparation derived from the individual serum;n n n Selectively separating the hyperimmune serum-reactive antigens and producing the hyperimmune serum-reactive antigens using chemical or recombinant methods. |
