Embodiments of the invention disclosed herein generally relate to anti-cocaine therapeutics. In particular, some embodiments of the present invention relate to a highly efficient, thermally stable, long-lasting cocaine esterase (CocE) mutant that can protect against the toxic and adjuvant effects of cocaine in a subject. Provided herein are mutant CocE polypeptides exhibiting heat stable esterase activity.
