abstract | The present invention provides a DNA molecule comprising i) a polypeptide of interest, and ii) a multicistronic transcriptional unit that encodes a selectable marker polypeptide acting in a eukaryotic host cell, wherein the polypeptide of interest is selected from a selectable marker polypeptide. Wherein the sequence encoding the polypeptide of interest is upstream from the coding sequence for the selectable marker polypeptide of the multicistronic transcriptional unit, wherein the internal ribosome entry site (IRES) is of interest A nucleic acid sequence downstream from the coding sequence for the polypeptide and upstream from the coding sequence for the selectable marker polypeptide, wherein the nucleic acid sequence encoding the selectable marker polypeptide in the coding strand comprises a GTG or TTG start codon. The invention also provides a method of obtaining a host cell expressing a polypeptide of interest, said host cell comprising a DNA molecule of the invention. The present invention further provides a method of producing a polypeptide of interest, said method comprising culturing a host cell comprising a DNA molecule according to the invention. |
