abstract |
The present invention relates to a floating cell line in which antisense lactic acid dehydrogenase mRNA is expressed and MnSOD is overexpressed to increase production of recombinant human EPO, and induces antisense lactate dehydrogenase (LDH) mRNA having a nucleotide sequence of SEQ ID NO: 1. The present invention provides a recombinant cell line which is transformed by a vector containing a manganese peroxygen-antimutant enzyme MnSOD gene having a gene having a nucleotide sequence of SEQ ID NO: 2 and a human erythropoietin with high efficiency. According to the above configuration of the present invention, the production of lactic acid accumulated in the culture medium is suppressed and the antioxidant capacity of the cell line is increased, thereby prolonging the life of the cell line, thereby increasing the EPO productivity. It is also advantageous to adapt adherent cells to floating cells by culturing in serum-free medium, having the ease of scale-up for mass production for industrialization. |
