The present invention relates to a filamentous phage display method wherein a desired polypeptide (POI) displayed on phage particles is cotranslated and translocated through the cytoplasmic membrane of a Gram-negative bacterium, based on a signal recognition particle pathway. . This method is particularly suitable when derived from polypeptides known to be difficult to display on phage, and proteins of cDNA libraries and other combinatorial libraries, especially very fast folding, stable protein scaffolds. The invention also relates to a phage or phagemid vector comprising a gene construct encoding a fusion polypeptide comprising a POI to be displayed on the phage particle and an N-terminal signal sequence that promotes co-translational translocation based on the signal recognition particle pathway.
