abstract A canola protein isolate is prepared having a protein content of at least about 90% by weight (Nx6.25), preferably at least about 100% by weight, consisting mainly of 2S canola protein and substantially free of 7S and 12S proteins. In one aspect, canola oil seed powder is extracted with an aqueous salt solution at elevated temperature to selectively extract 2S canola protein from wheat to produce a canola protein extract solution containing predominantly 2S protein. 2S canola protein is recovered as an isolate. In another aspect, the canola oil seed powder is initially extracted with water to preferentially extract 7S and 12S canola proteins, and then the canola oil seed powder is extracted with an aqueous salt solution to extract 2S protein from wheat. 2S canola protein isolate is obtained from the salt extract. In another aspect, canola oil seed flour is extracted from an aqueous salt solution to extract 2S, 7S and 12S proteins from wheat. The aqueous protein extract solution can be heat treated at elevated temperature to precipitate 7S and 12S proteins and leave 2S protein solution from which the isolate can be recovered. In another aspect, the aqueous protein solution is concentrated prior to heat treatment.
