titleOfInvention | Compositions and Methods for Metabolic Screening of Transfected Cells |
abstract | The present invention relates to novel selection marker vectors and methods of using these vectors to build stable gene expression systems in eukaryotic cells. More specifically, the present invention provides compositions and methods that use any enzyme useful in the eukaryotic sterol / cholesterol biosynthetic pathway, such as 3-ketosteroid reductase, as a metabolic selection marker for selecting transfected cells. In one embodiment, the method has the ability to transfect cells that are nutritionally constitutive to cholesterol with a vector encoding a 3-ketosteroid reductase and one or more heterologous proteins and / or have the ability to survive in a cholesterol deficient medium. And / or selecting cells that produce heterologous proteins in said cells in serum-free and / or serum-free medium. |
