abstract | The present invention relates to a method for detecting neuronal hyperactivity such as seizures and epilepsy by measuring the expression level of calcium-binding protein (CBP) in specific nerve cells of the central nervous system, including parvalbumin or calbindin D. Calcium-binding protein (CBP) is a useful marker for certain neurons in the central nervous system, but it is unclear whether changes in CBP expression in the hippocampus actually indicate neuronal activity. Therefore, in the present invention, the expression of CBP expression in the rat hippocampus was given to give an electric shock (ECS), and as a result, the expression of CBP in the hippocampus was decreased in proportion to the degree of stimulation after 6 hours after the stimulation. Restored to its original state. However, the paired-pulse response of the dentate gyrus was temporarily reduced after electroconvulsive shock and soon normalized to baseline. In addition, the effects of the seizure shock on the expression of CBP and the twin-pulse reaction could be prevented by pre-treatment with vivathrine. These findings indicate that decreased expression of calcium-binding protein due to seizures indicates hyperactivity of CBP-positive neurons, which is the result of abnormal discharge, and thus replaces the current behavioral ECS assays. New anticonvulsant assays have been made that make the present invention applicable. |
