Cold homogeneous detection during or after asymmetric PCR amplification, preferably LATE-PCR, using fluorescent DNA dyes and indirectly excitable labeled mismatch-allowing probes allows analysis of the complex target. Sequencing sample preparation methods after LATE-PCR amplification reduce complexity and allow for a "single in vitro" process.
