abstract The present invention provides a polynucleotide which has a high RNA interference effect on a target gene and has a very low risk of RNA interference on a gene not related to the target gene. From the base sequence of the target gene of RNA interference, the sequence site | part which follows the rules of (a)-(d) is searched, and the polynucleotide which produces RNAi based on a search result is designed and synthesize | combined. (a) The base at the 3 'end is adenine, thymine or uracil. (b) The base at the 5 'end is guanine or cytosine. (c) The 7-base sequence of the 3 'end is rich in one or two or more bases selected from the group consisting of adenine, thymine and uracil. (d) The number of bases that can cause RNA interference so as not to cause cytotoxicity.
