abstract A method and kit for use in detecting any one of multiple or specific mutations, in particular multiple chromosomal abnormalities resulting from microdeletion, in a single assay. Polymerase chain reaction (PCR) is performed to amplify eukaryotic genomic DNA using a plurality of primer oligonucleotide pairs, with one primer in each pair having a detectable label attached to its five ends. Multiple primer pairs target DNA fragments of those different chromosomes that exhibit potential chromosomal abnormalities, and one pair targets control genes. The amplified PCR product is purified to obtain single-stranded DNA with a detectable label from which the single-stranded DNA comprises a DNA oligonucleotide probe having a nucleotide sequence complementary to one strand of nucleotide sequence of each fragment. Hybridized to points on each containing microarray. This microarray is imaged for the presence of a label at each point thereof, first comparing the results of this image with an image of a point specific to the control gene, and then comparing the results obtained from imaging of normal DNA to the target of interest.
