Methods and compositions for reducing nonspecific off target effects in RNA interference are provided. The methods and compositions provide an effective and efficient application of RNA interference in applications of the present invention, such as diagnostics and therapeutics, through modifications to siRNA. More specifically, modified siRNAs with reduced off-target effects occurring in the gene-silencing process have been developed. Modifications include 2'-O-alkyl or mismatch modifications at specific positions of sense and / or antisense strands. |
